Personally, I prefer a matrix as done in many other games, not colored lines. That's how you could fit in 16 civs and their relationships.
16 civs on the current format would certainly be messy. Let's hope they've done something nifty.

Firaxis stated a while ago that 16 civs will not be supported by many screens and will only be available by using the editor. That's enough for me though, as long as I have my 16 civs...

Yeah, but three days ago Firaxis stated that 16 civs are default for huge worlds. I guess that means 'supported by all screens'.

IMO, there are at least two good reasons for Firaxis´change of mind:

        - the need to jolly the fans along while delaying MP,
        - the 180 playable nations of EU2.

Hmm. You´re right. Especially displaying several military alliances with and against other civs could be tricky.

I can't wait to play the first 16 civ megamap World War II scenario.

Let me think how I would fill up those slots:

              The Big Five

              USA
              Britain
              Nazi Germany
              Imperial Japan
              USSR

              The Little Powers

              Nationalist China
              Communist China
              Finland
              Fascist Spain
              Turkey
              Fascist Italy
              Peronist Argentina
              South America
              Canada
              Australia/NZ
              British India

Imagine the possiblities....what if the Finnish government had become Communist?
What if India had revolted against Britain?
What if Argentina had become militarily aggressive in South America?
              "Wait a minute..this isn''t FAUX dive, it's just a DIVE!"
"...Mangy dog staggering about, looking vainly for a place to die."
"sauna stories? There are no 'sauna stories'.. I mean.. sauna is sauna. You do by the laws of sauna." -P.

I dont think default means supported by all screens. Only time will tell.

Things that Firaxis actually has to modify:

                  - the foreign advisor screen (somewhat tricky),
                  - the histograph screen (fairly easy),
                  - the civ colours (and heck, even CtP2 managed to display 16 distinguishable colours).

Doesn´t sound utterly impossible to me. And again: I believe that Firaxis needed to include the '16 civs'-feature because EU2 (with 180 playable nations) will be released shortly after Civ3. (And while EU can be tedious, '100 nations' means very interesting diplomacy!)

If it's available in the new game options, it would have to be supported by in-game screens.
