You should always try to get an army up ASAP (unless you've chosen to play with peace-time, which might be a good idea, until you've learned the game)
Always try to max out you income in the different resources, but don't go over the limit, as it would just be wasted, and research commerce to get the limit higher. It's not always the best idea to research to the next age asap, often there are more important research to be done
In general, build a diverse army of different unit types. This game favors a "rock paper scissors" power relationship, so no type of unit is truly dominant. Get to know which types of units are best for countering others (this is in the manual and also in the in-game help).
Like Adagio said, max out your resource collection, but don't allow Citizens to collect a resource that's already maxed out, since that's a waste of Citizens (and by extension, a waste of the Food you used to build the Citizens). Mouseover a maxed-out resource in the upper left corner of the screen, and you'll see an "efficiency" rating - try to keep that at 95% or higher. (i.e. remove citizens from work until that rises to 95%+)
Research the booster techs at the Granary, Lumber Mill, University, Temple, and Smelter so you get more resources per collecting Citizen.
In my opinion, you should always keep your Science research (yellow) at parity with your other research, since that line of research actually decreases the cost of others. This may become less important later in the game. However, the notable exceptions to this case are Civics (blue) if you really need to build another city, and Commerce (green) if your resource limits are way too low.
If your'e having difficulty warding off the computer's attacks, remember that the AI handles attacks poorly. If you send an army over to attack one of its cities, it will pretty much recall all its units and try to defend that threat.
I've found the easiest thing to do is just alternate one of each unit. i.e. LI/HI/Archer/LI/HI/Archer. Takes no smarts, get the cheap ramp-up cost. As you get better you learn other ways.
