My choices were limited for half hour shows because I only really watch 4 non-animated half hour shows... Malcolm in the Middle, Scrubs, My Name Is Earl, and Office.
There really aren't a lot of good female characters in those shows. I mean, there are, but not any AWESOME ones. I'm not talking about looks, but rather, characters that define the show. Most of the shows, in fact, are carried by the casts as a whole, so it's hard to pick just one member who stands apart from the rest. My Sarah Chalke pick is more just a reflection of how good the Scrubs cast is.
Another thing I'd like to see...

There really aren't a lot of good female characters in those shows. I mean, there are, but not any AWESOME ones. I'm not talking about looks, but rather, characters that define the show.
Yeah, I realized that as a problem. It seems that the awesome roles are given to the males in most half hour shows. Most female roles are foils for the male roles it seems.
Elliot in Scrubs was the one I picked because of the zaniness of the character. She's like a female J.D. Many more juicy roles for women in hour long shows.“I give you a new commandment, that you love one another. Just as I have loved you, you also should love one another. I just used yours
I mean, Chloe from 24... and Starbuck are great...
but still...
compared to like Jack Bauer and Vic Mackey from The Shield? Where are the awesome female characters? It doesn't seem like there really are any.
There are good ones in supporting roles, but none that make me say ZOMFG "JACK BAUER" you know?
Although Chloe did cap some mofos last seasonTo us, it is the BEAST.

I don't watch much TV. As far as English-language shows go, I pretty much only watch Battlestar Galactica, Desperate Housewives, Lost, Rome (which had its finale here last weekend) and Mythbusters -- and if I'm at home reruns of ER, 'cuz there's not really anything else worth watching at dinner time and I like to have some background noise when I'm cooking/eating. Oh, and of course BBC Grandstand, best television on earth

Very true. I think "Alias" may be on the air (has it been canceled), but Jennifer Garner's chacter in past years was pretty cool. Lately the show, and her character, have sucked.
Jessica Alba in "Dark Angel" was good as well. I never really watched "Buffy the Vampire Slayer", so I don't know how awesome that character was.

Speaking of those forensic detective shows (or things like them), why do they all have male leads? Grissom for CSI, Horatio Assface for CSI:Miami, That dude whose been in those movies for CSI: NY, Mark Harmon in NCIS, etc.

Best One Hour TV Show: The only show I really look forward to anymore is Desperate Housewives and Grey's Anatomy... and Las Vegas, but then that's just for the boobs

well, the lead lab specialist in NCIS is female... "Abby"
she handles all the crime scene stuff
although the lead doctor is a dude... he does the autopsy stuff
but Abby usually does everything else... Gibbs (Mark Harmon) is the lead agent, but he doesn't handle crime scene stuff at all
It's not really a show that is focused on crime scene stuff, although it is part of the show.To us, it is the BEAST.
