I believe that we should set forth to build cities at aproximately the #5 & #6 sites after Pampalona is finished. Frankly, I would like to see the site for #5 one square to the east. This site would overlap with #6, but it would be well defended with the river to the north protecting it. We need to establish our control over this valley before ND gets bottled up in the north and has to slough through the jungle to expand. We also need to prepare for the eventual war and a city with walls behind a river has a far greater defense.
We would then add another site on the coast to the SW of new #5.
I'm sure we all like having little-to-no overlap, but in this game against humans, positioning, territory, and numbers are more important than expected growth in the modern ages. I believe we should take advantage of hills and rivers as much as possible.
As to the question of "should we backfill?" I believe that we will to a certain degree. If it turns out that there are no neighbors to the immediate west we should focus all of our early expansion efforts into Cow Valley and then move our settlements to the south-west and north-east after that.
