You may want to reinstall the game. New CD drives shouldn't have a problem unless its bad drive. However if you have ADDED the drive rather than replaced it your drive letters are now different in any case. For some reason (carelessness I would guess) Civ III has trouble running from a different drive than it was installed from.
What I would do is copy the save folder and any other mods you have to another place on the drive. Unistall and then reinstall.
Actually I would try hacking the registry to chang the drive the game wants to run from myself but I can't recommend that unless you are experienced in it. Reinstalling should do the trick unless the drive itself has a problem.

Another alternative since you own the game' is to pick up the hacked .exe that doesnt require the CD. Thats what I did when a program I installed added a virtual drive with a drive letter before my cd drives. It took me ten minutes to find and install it. Why do they bother with the bull**** copy protection?We need seperate human-only games for MP/PBEM that dont include the over-simplifications required to have a good AI
