There is a definite memory leak in this game, and as you play it will slow down dramatically, especially on large maps. I have also read rumors that it might be ADM64 related (which I have) but I am not sure on that.
I have 1 gb of memory, and Civ has twice slowed down significantly and eventually crashed and my XP has told me that it was increasing the Page Cache because it had run out of memory. That is insane. If I close it down, reboot and start again, everything is fine again.
